Forschung
Biologische Membranen sind für das Leben unerlässlich. Sie stellen spezialisierte Permeabilitätsbarrieren für Zellen und Zellorganellen dar, in denen das Zusammenspiel von Lipiden und Membranproteinen grundlegende Prozesse der Energieumwandlung, des Transports von Proteinen und gelösten Stoffen, der Signalwahrnehmung und -weiterleitung sowie der Motilität ermöglicht. Hauptverantwortlich für die Funktion sind Membranproteine, die auch die meisten Angriffspunkte für Arzneimittel darstellen. Proteinstrukturen sind die Grundlage für ein vollständiges Verständnis des Wirkmechanismus und für die Aufklärung der Ligandenbindung. Dennoch sind die Strukturen von Membranproteinen, insbesondere von eukaryotischen und menschlichen Membranproteinen, immer noch selten. Darüber hinaus ist wenig über die Wechselwirkung mit Lipiden und intrazellulären Signalwegen bekannt, die für die Funktion entscheidend sind.
Die Forschungsarbeiten der Gruppe konzentrieren sich auf Membranproteine, die eine wichtige Rolle bei der biologischen Energieumwandlung und dem Ionentransport spielen. Untersucht werden Struktur, Mechanismus und Funktion von respiratorischen Mehreinheitskomplexen und von Ionentransportern sowie deren Interaktion mit intrazellulären Signalwegen. Der experimentelle Ansatz kombiniert molekularbiologische, biochemische und biophysikalische Methoden mit Schwerpunkt auf der Röntgenkristallographie zur Strukturbestimmung.
